Inhibition of casein kinase 2 blocks G2/M transition in early embryo mitosis but not in oocyte meiosis in mouse

چکیده
Casein kinase 2 (CK2) is a highly conserved, ubiquitously expressed serine/threonine protein kinase with hundreds of substrates. The role of CK2 in the G2/M transition of oocytes, zygotes, and 2-cell embryos was studied in mouse by enzyme activity inhibition using the specific inhibitor 4, 5, 6, 7-tetrabromobenzotriazole (TBB). Zygotes and 2-cell embryos were arrested at G2 phase by TBB treatment, and DNA damage was increased in the female pronucleus of arrested zygotes. Further developmental ability of arrested zygotes was reduced, but that of arrested 2-cell embryos was not affected after releasing from inhibition. By contrast, the G2/M transition in oocytes was not affected by TBB. These results indicate that CK2 activity is essential for mitotic G2/M transition in early embryos but not for meiotic G2/M transition in oocytes.

A casein kinase I isoform is required for proper cell cycle progression in the fertilized mouse oocyte.
Casein kinase I is a family of serine/threonine protein kinases common to all eukaryotes. In yeast, casein kinase I homologues have been linked to the regulation of growth, DNA repair and cell division. In addition, their subcellular localization to membraneous structures and the nucleus is essential for function. In higher eukaryotes, there exist seven genetically distinct isoforms: (alpha), s...
متن کاملP-90: The Effect of Nitric Oxide on Mouse Oocyte in Vitro Maturation in Two and Three Dimensional Conditions
Background: In vitro culture of ovarian follicles may preserve fertility in women with premature ovarian failure due to cancer .It seems that creation a condition that could maintain cellular communications and supports growth of follicles to produce mature oocytes appear to be essential. Nitric oxide (NO) has been recently shown to act with a dual action in mouse oocyte meiotic maturation depe...
متن کاملCasein Kinase 1 Alpha Regulates Chromosome Congression and Separation during Mouse Oocyte Meiotic Maturation and Early Embryo Development
Casein kinase I alpha (CK1α) is a member of serine/threonine protein kinase, generally present in all eukaryotes. In mammals, CK1α regulates the transition from interphase to metaphase in mitosis. However, little is known about its role in meiosis. Here we examined Ck1α mRNA and protein expression, as well as its subcellular localization in mouse oocytes from germinal vesicle to the late 1-cell...
متن کاملChk2 Regulates Cell Cycle Progression during Mouse Oocyte Maturation and Early Embryo Development
As a tumor suppressor homologue during mitosis, Chk2 is involved in replication checkpoints, DNA repair, and cell cycle arrest, although its functions during mouse oocyte meiosis and early embryo development remain uncertain. We investigated the functions of Chk2 during mouse oocyte maturation and early embryo development. Chk2 exhibited a dynamic localization pattern; Chk2 expression was restr...
